FUJIMURA, Y., KAWAMURA, M. and NARUSE, H. Simultaneous Quantitative Estimation of Galactose-1-Phosphate and Galactovve in Blood for the Diagnosis of Galactosemia. Tohoku J. exp. Med., 1982, 137 (3), 28°-295 A new microfluorometrical simultaneous assay method of galactose-l-phosphate and galactose in blood discs was devised by use of alkaline phosphatase and fl-galactose dehydrogenase. Our method statistically corresponded well with the Kirkman's method. It can detect 1>< 10-10 mole of minimal concentration of galactose-lphosphate and galactose in one blood disc paper (3 mm in diameter), and this means the sensitivity of assay of galactose-l-phosphate was 0.1 mg%. Assay range in our method was very broad (0-2 mM or 0-10 mM). The accuracy and reproducibility of galactose-l-phosphate assay were 3.4+0.1 mg%, 8.0±0.4 mg% or 15.1±0.6 mg%. Mean values of galactose-l-phosphate and galactose in blood of normal infants were 0.8 mg% and 0.3 mg%, respectively. We applied this method to mass screening of galactosemia and could accurately distinguish many positive and false positive cases detected by Paigen's and Beutler's methods. This method gave us an easy and accurate assay system for the diagnosis of uridyl transferase and galactokinase deficiencies. -galactosemia; galactose-l-phosphate; galactose; mass screening of galactosemia

Quantitative Bacterial Micro-Assay for Rapid Diagnosis of Galactosemia: Application in Galactosemia Neonatal Screening

In the present study a new economic and rapid bacterial micro-assay for simultaneous detection and quantitative measurement of serum galactose was developed. Analysis of the standard curve showed a linearity range for galactose from 2 mg/dL to 180 mg/dL with a regression equation of Y = 0.013X ? 0.083; R² = 0.962. The advantage of the method is its ability to measure serum galactose quantitativ...

Problems in the diagnosis of transferase and galactokinase deficient galactosemia.

Galactose in serum and galactose-1-phosphate in erythrocytes were measured in six transferase deficient children to determine if these metabolites could be used in detecting transferase deficient galactosemia. In all six children the galactose levels were normal and the galactose-1-phosphate elevated. The galactose level depends on diet and the rate of metabolism to galactose-1-phosphate and, t...

Quantitative Beutler test for newborn mass screening of galactosemia using a fluorometric microplate reader.

BACKGROUND The Beutler enzyme spot test is an effective assay for newborn mass screening of galactosemia, but it is qualitative and relies on visual interpretation. We describe a quantitative, instrumental modification of the assay. METHODS We modified the macroscopic visual Beutler enzyme spot test by adding extraction of blood components from filter paper, deproteinization with acetone-meth...

Rapid method for screening for galactosemia and galactokinase deficiency by measuring galactose in whole blood spotted on paper.

Two major metabolic disorders are associated with galactose metabolism: galactokinase (ATP : D-galactose1-phosphotransferase, EC 2.7.1.6) deficiency and galactosemia (1) secondary to a deficiency in galactose-i-phosphate uridyl transferase (UTP: a-D-galactose-i-phosphate uridyltransferase, EC 2.7.7.10) activity. One case reported in 1972 (2) described a third enzymatic (UDPglucose 4-epimerase, ...
